چکیده

Matrix metalloproteinases are an integral component of the organ-specific tissue microenvironment, primarily participating in regulation state integrative-buffer metabolic environment, remodeling, morphogenesis and immunogenesis, genesis pathological vascular diseases. Therefore, issues related to matrix metalloproteinase activity under altered gravity a matter high urgency for space biomedicine. The review highlights aspects gravity. study results included current systematic were searched independent literature sources PubMed Cochrane databases, Google Scholar NASA Technical Reports Server. A variety stress factors associated with flights, particular, exposure radiation, enhances expression transforming growth factor beta metalloproteinases-2. Experimental data obtained during several flights demonstrate that microgravity affects architecture cells increases metalloproteinases-1 interleukin-6. Microgravity reduces collagen I, level fibrillar collagen, which mechanical properties intracellular matrix. -1, -3, -10 demonstrated increased flight samples 16-week-old female C57BL/6J mice (n = 8) 15 days aboard Space Shuttle Discovery, STS-131 mission, compared ground control (12.94; 2.98 16.85 times, respectively, P<0.05), while inhibitors – inhibitors-1, -2, no statistically significant changes gene expression. There determined differences profiles lungs animals groups. genes revealed more than doubled CTGF, MMP-2, NACM1, SPARC, SPOCK1 TIMP-3, greatest decrease LAMA1, MMP-3, MMP-7, MMP-13, VCAM-1, SELE.

Matrix metalloproteinases and their inhibitors in connective tissue remodeling.

Matrix metalloproteinases are an important group of zinc enzymes responsible for degradation of the extracellular matrix components such as collagen and proteoglycans in normal embryogenesis and remodeling and in many disease processes such as arthritis, cancer, periodontitis, and osteoporosis. A matrixin family is defined, comprising at least seven members that range in size from Mr 28,000 to ...

Extracellular matrix remodeling and matrix metalloproteinases in the vascular wall during aging and in pathological conditions.

The extracellular matrix provides a structural framework essential for the functional properties of vessel walls. The three dimensional organization of the extracellular matrix molecules - elastin, collagens, proteoglycans and structural glycoproteins - synthesized during fetal development - is optimal for these functions. In uninjured arteries and veins, some proteases are constitutively expre...

Ovarian tissue remodeling: role of matrix metalloproteinases and their inhibitors.

Follicular formation, growth or atresia, and ovulation as well as luteal formation and subsequent regression are dependent upon cyclical remodeling of the extracellular matrix (ECM). The proteinaceous and nonproteinaceous components of the ECM provide the tissue specific, extracellular architecture to which cells attach. Furthermore, the ECM modulates cellular activities through cellular surfac...

Molecular imaging of activated matrix metalloproteinases in vascular remodeling.

BACKGROUND Matrix metalloproteinase (MMP) activation plays a key role in vascular remodeling. RP782 is a novel indium (111)In-labeled tracer with specificity for activated MMPs. We hypothesized that RP782 can detect injury-induced vascular remodeling in vivo. METHODS AND RESULTS Left common carotid artery injury was induced with a guidewire in apolipoprotein E(-/-) mice. Sham surgery was perf...

Biglycan fragmentation in pathologies associated with extracellular matrix remodeling by matrix metalloproteinases

BACKGROUND The proteoglycan biglycan (BGN) is involved in collagen fibril assembly and its fragmentation is likely to be associated with collagen turnover during the pathogenesis of diseases which involve dysregulated extracellular matrix remodeling (ECMR), such as rheumatoid arthritis (RA) and liver fibrosis. The scope of the present study was to develop a novel enzyme-linked immunosorbent ass...
